My son was a full time student and also worked for 4 months in 2021, and qualifies under IRS checklist to be claimed as dependent. However, it is more advantageous for both returns for me not to claim him, and thus let him qualify for additional tax benefits given his low income. However, there is check box “someone can claim me as a dependent” in 1040, and if checked disqualifies him. Does that box need to be checked even if I do not claim him as a dependent? Thanks!

Unfortunately, the tax benefits your son might otherwise qualify to receive are available only for those who cannot be claimed as a dependent. You can’t make those benefits available to him by making the voluntary choice not to claim him as a dependent. This is the reason for the checkbox.
